Production of recombinante laccase

This project aims at the heterologeous expression of the Melanocarpus albomyces laccase gene in Trichoderma reesei in fed-batch culture with special emphasis on the volumetric productivity of the cultivation. Different feeding strategies will be applied in order to maximize the space-time-yield of recombinant enzyme formation. As the promotor used in the recombinant strain is obtained from a cellulase gene of Trichoderma, the hypothesis to be prooven is, that protein expression will be high under conditions of low specific growth rates.
