During a predecessor project with TU Vienna, the Rhamnose-induced expression of the product alkaline phosphatase has been characterized. Here, the C-source supply was identified as a main impact on specific productivity (qP). As a main outcome of these investigations, the different parameters, which determine the C-source feed (and, thus, the productivity) during cultivation, could be condensed to one physiological parameter: The specific substrate uptake rate (qS).
The current project described in this annex, is a follow-up of the aforementioned work. The concept identified in the predecessor project, should be verified with two more marked-relevant products. The relation of the specific substrate uptake rate and the specific productivity will be characterized. By establishing this relationship, a universal approach for speeding-up process development will be defined, and ideally a universal platform property can be identified. Moreover, establishing a control strategy on the specific substrate supply could increase the fermentation productivity.
