Learning outcomes

After successful completion of the course, students are able to grow a microorganism for enzyme production, to purify the produced enzyme and characterize it kinetically. In addition, the students will be able to characterize the microbial production host by molecularbiological methods.

Subject of course

Sterile handling and basic microbiological techniques. Recombinant production of an enzyme in fermentation. Purification and kinetic characterization of the enzyme. Analysis of the microbial production strain by molecuar biology methods (PCR).

Teaching methods

The contents of the laboratory course are conveyed by a working instruction and the experiments described therein. A deepening of the teaching content is achieved by the preparation of a laboratory protocol by the students. In addition, open questions are addressed through preparation, results and protocol discussions.

Registration for the courses will take place from 14 August 9.00 via TISS. Please register directly into one of the groups.

The prerequisite for registration is the positive completion of the VO Biochemistry I. If the number of registrations exceeds the number of available laboratory places and it is therefore necessary to rank the registered students with fulfilled entry requirements, the following ranking criteria apply:


Ranking criterion 1: Positive passing of the exam Mathematik für TCH I (104.084). Date and grade of this exam is NOT a criterion.

Ranking criterion 2: Date of the positively passed VO Biochemistry I examination (172.695)

Ranking criterion 3: Grade of the examination VO Biochemistry I (172,695)

Ranking criterion 4: Date of registration for the laboratory exercise

Based on last year's experience, it is unlikely that it is necessary to rank registered students with fulfilled entry requirements. And all students can register directly in TISS for one of the courses.

People taking the VO Biochemistry I exam in the winter semester can still register on a space-available basis.

For all courses there will be a compulsory preliminary meeting. The preliminary meeting will take place online in Zoom. Detailed information will follow end of September.

A written test will take place before the courses start. Exact dates for the individual course exams will be available in TISS from the end of September. Content is theory and execution of the exercises as stated in the lab course instructions (Arbeitsvorschrift). The instructions will be published middle of September in TISS.

The course takes place in the student laboratory in the BH building on the ground floor. The procedure and details can be found in TISS from middle of September on.

Examination modalities

Written exam at the beginning of the course. Performance within the lab course. Delivery of the lab protocol. The cooperation is included in the assessment of the laboratory protocol. The written examination lasts 30 minutes and consists of 6 questions, which are assessed with one point.  ≥ 5.0 points correspond to a "Very Good", ≥ 4.3 points to a "Good", ≥ 3.6 points to a "Satisfactory", ≥ 3.0 points to a "Sufficient" and less than 3.0 points to a "Not Sufficient". If the written examination is negative, it can be repeated once more before the start of the course.

The grade of the laboratory exercise results from the partial grades for the introductory test and the protocol with a weighting of 40:60.

The theoretical background can be retrieved from the basic lectures "Biochemistry 1", "Biochemistry Proseminar" and "Introduction to Biotechnology and Bioengineering"

Previous knowledge

The positive completion of the exam "Biochemistry I" is an essential prerequisite for registration to the course. To attend the course the positive passing of a written exam about the content and background of the individual course examples is a prerequiste.

We further strongly recommend also to complete the exam on "Introduction to Biotechnology and Bioengineering" and "Biochemistry Seminar" before this lab. This will give you a deeper understanding of the course and help to pass the exam positively.
