After successful completion of the course, students are able to grow a microorganism for enzyme production, to purify the produced enzyme and characterize it kinetically. In addition, the students will be able to characterize the microbial production host by molecularbiological methods.
Sterile handling and basic microbiological techniques. Recombinant production of an enzyme in fermentation. Purification and kinetic characterization of the enzyme. Analysis of the microbial production strain by molecuar biology methods (PCR).
The contents of the laboratory course are conveyed by a working instruction and the experiments described therein. A deepening of the teaching content is achieved by the preparation of a laboratory protocol by the students. In addition, open questions are addressed through preparation, results and protocol discussions.
Registration for the courses will take place with the beginning of September via TISS/TUWEL. Please register directly into one of the different groups. The prerequisite for registration is the positive completion of the VO Biochemistry I. If the number of registrations exceeds the number of available laboratory places and thus a ranking of the registered students with fulfilled entrance requirements becomes necessary, the following ranking criteria apply:Ranking criterion 1: Positive passing of the exam Mathematik für TCH I (104.084). Date and grade of this exam is NOT a criterion.Ranking criterion 2: Date of the VO Biochemistry I examination (172.695)Ranking criterion 3: Grade of the examination VO Biochemistry I (172,695)Ranking criterion 4: Date of registration for the laboratory exerciseAs it is probably necessary to rank the registered students with fulfilled entrance requirements, the registration will be carried out according to the following procedure:All students who have already completed the Mathematics for Technical Chemistry I AND the VO Biochemistry I can register directly at TISS/TUWEL.All students who have not yet completed Mathematics for Technical Chemistry I, but who have passed the VO Biochemistry I, cannot register directly via TISS/TUWEL. They have to send me their certificate of Biochemistry I by email from the 15th of September on (bernhard.seiboth@tuwien.ac.at), so that I can make a ranking. Please indicate in the email your desired course + all possible alternative courses. The course allocation for students who have only completed VO Biochemistry 1 will take place end of September.For all courses there will be a compulsory preliminary meeting. The preliminary meeting will take place online. Detailed information will follow end of September.A written test will take place before the courses start. Exact dates for the individual course exams will be available in TISS from the end of September. Content is theory and execution of the exercises as stated in the Arbeitsvorschrift. The positive completion of the exam "Biochemistry I" is an essential prerequisite for registration to the course. To attend the course the positive passing of a written exam about the content and background of the individual course examples is a prerequiste.
We further strongly recommend also to complete the exam on "Introduction to Biotechnology and Bioengineering" and "Biochemistry Seminar" before this lab. This will give you a deeper understanding of the course and help to pass the exam positively.
