Learning outcomes

After successful completion of the course, students are able to describe and explain the following topics:

  • Basis of instrumental analysis: signals (types, filter methods), unwanted influencing of signals (noise, drift and interference) and methods for improving the signal / noise ratio.
  • The theory and basics of chromatographic separation techniques in the liquid and gas phase. This includes the principle of distribution on which the separation technology is based, the terms selectivity, efficiency and resolution and their various forms of implementation (gas chromatography, thin-layer chromatography, HPLC, IC, SEC, affinity chromatography) with the corresponding device technology and relevant application examples.
  • The basic principle and the technical implementation of the electrophoretic separation techniques gel and capillary electrophoresis (GE, CZE), isoelectric focusing (IEF) and isotachophoresis (ITP))
  • The basics and realizations of important electroanalytical methods (conductometry, potentiometry, amperometry, coulometry).
  • The measuring principles, the instrumental implementation and areas of application of the spectroscopic methods in the areas of element analysis (atomic absorption and atomic emission spectrometry, X-ray fluorescence analysis) and molecular analysis (UV / Vis absorption spectroscopy, fluorescence spectroscopy, IR spectroscopy, chemical and bioluminescence).

 

Subject of course

  • Theoretical basics and practical implementation of the chromatographic separation techniques in liquid and gas phase (gas chromatography: GC, liquid chromatography: HPLC, thin layer chromatography: DC; ion chromatography: IC; size exclusion chromatography: SEC; bioaffinity chromatography)
  • Theory and practice of electrophoretic separation techniques
  • Scaling of separation processes (miniaturization / scale-up)
  • Fundamentals and implementation of important electroanalytical processes
  • Basics of processing analytical signals (signals and noise)
  • Calibration of analytical processes
  • Basics and concept of modern spectroscopic methods:
  • Elemental analysis: AAS and OES
  • Molecular spectroscopy:
    - UV / Vis spectroscopy
    - infrared spectroscopy
    - Raman spectroscopy
    - fluorescence spectroscopy

Teaching methods

The course will be held in the classical lecture format (via LectureTube), supported by the presentation of PowerPoint slides. In order to support understanding, and also to be able to answer potential questions, Q&A sessions (via ZOOM) will be held in regular intervals.

Examination modalities

Written exam with currently four questions that come from the two parts of the lecture course. To successfully pass theexam, at least 50% of the total number of points (currently: 40 points) are required. The key to the grades is:

  • 40,0 – 35,0 points: Very good
  • 34,5 – 30,0 points: Good
  • 29,5 – 25,0 points: Satisfactory
  • 24,5 – 20,0 points: Sufficient
  • <20,0 points: failed.

The exams for the current lecture course take place - if not mentioned otherwise - on the Fridays listed below at 14.00-16.00 h in the Radinger lecture hall.
